چکیده

An optimization algorithm was used to compute parameter values for kinetic models of NH4 transport by the renal Na-K-2Cl cotransporter (NKCC2) and K-Cl cotransporter (KCC). The optimization method finds model off-binding and translocation rate constants by minimizing the difference between model unidirectional fluxes and published data on NH4 -dependent 86Rb+ uptake by the NKCC and KCC transporters in transfected Xenopus oocytes. The computed parameters for each model yielded fluxes consistent with the experimental curves. However, a systematic exploration of the parameter space revealed that, in some models, different parameter sets yield essentially identical fits to the data. With the identified parameters, the NKCC models show K/NH4 exchange and enhancement of Na + uptake by NH4 binding when external K + concentration is low. Further, under these conditions, substantial rates of K/NH4 exchange are predicted, which may prevent depletion of luminal K+. For the KCC models, in the medulla where the interstitial concentration of ammonium is high, substantial rates of K/NH4 exchange are again predicted to occur.

Ammonium transport and pH regulation by K(+)-Cl(-) cotransporters.

The Na(+)-K(+)-Cl(-) cotransporters (NKCCs), which belong to the cation-Cl(-) cotransporter (CCC) family, are able to translocate NH4(+) across cell membranes. In this study, we have used the oocyte expression system to determine whether the K(+)-Cl(-) cotransporters (KCCs) can also transport NH4(+) and whether they play a role in pH regulation. Our results demonstrate that all of the CCCs exam...

Functional significance of channels and transporters expressed in the inner ear and kidney.

A number of ion channels and transporters are expressed in both the inner ear and kidney. In the inner ear, K(+) cycling and endolymphatic K(+), Na(+), Ca(2+), and pH homeostasis are critical for normal organ function. Ion channels and transporters involved in K(+) cycling include K(+) channels, Na(+)-2Cl(-)-K(+) cotransporter, Na(+)/K(+)-ATPase, Cl(-) channels, connexins, and K(+)/Cl(-) cotran...

A possible catalytic role for NH4+ in Na+ reabsorption across the thick ascending limb.

ACTIVE REABSORPTION of Na across the thick ascending limb of Henle (TALH) plays an essential role in the urine concentrating mechanism, as it generates and maintains the axial osmolality gradient in the outer medulla. Na enters the TALH cell mostly through apical Na -K -2Cl (NKCC2) cotransporters, to a lesser extent through Na /H (NHE3) exchangers, and it is extruded at the basolateral membrane...

Transport and regulatory properties of the apical Na-K-2Cl cotransporter of macula densa cells.

[Formula: see text]/NH3fluxes were used to probe apical Na-K-2Cl transport activity of macula densa (MD) cells from rabbit kidney. In the presence of 25 mM NaCl and 5 mM Ba2+, addition of 20 mM[Formula: see text] to the lumen produced a profound intracellular acidification, and ∼80% of the initial acidification rate was bumetanide sensitive. The[Formula: see text]-induced acidification rate was...

A mathematical model of rat ascending Henle limb. III. Tubular function.

K+ plays a catalytic role in AHL Na+ reabsorption via Na+-K+-2Cl- cotransporter (NKCC2), recycling across luminal K+ channels, so that luminal K+ is not depleted. Based on models of the ascending Henle limb (AHL) epithelium, it has been hypothesized that NH4+ may also catalyze luminal Na+ uptake. This hypothesis requires that luminal NH4+ not be depleted, implying replenishment via either direc...
